Not sure the whirly thing is actually supposed to be a hole in space, but I can't not vote for that Marie Severin (looked it up) Captain Marvel cover. Scott Harris was a very close 2nd for me.

This was a tough one to find the cover that best hit the mark. I agree with berk, that it is uncertain if the galaxy spiral on the Captain Marvel cover contains a black hole or worm hole (and I have a copy of it) as the theme warranted, but it is definitely cool on an interstellar scale so Pinkfloydsound17 gets my vote.
Ishkabibble is very close second.
