That animated Spectre short written by Steve Niles is excellent. Jim Corrigan also appears in the first episode of Constantine "Danse Vaudou".

The Flash, if done right, could make for an amazing movie. A Deadman movie might also be very interesting.

Amethyst Princess of Gemworld could make an interesting live action series or film. DC Nation actually did pretty cool series of animated shorts.

The Spectre and Jonah Hex, as well as a PROPER Punisher movie. I'd love to see a Spectre movie too, especially if it was done like this: Yeah that short is fantastic. That and the Jonah Hex ones both captured the EXACT approach needed for the silver screen for those characters and in the case of The Spectre whats needed for the comics as well (Jonah Hex of course already had an amazing modern run).

Amethyst Princess of Gemworld could make an interesting live action series or film. DC Nation actually did pretty cool series of animated shorts. I'm still waiting for Amethyst to land a regular animated series and a kid-friendly ongoing comic series. Too much potential for an all-ages following that's just being wasted right now.

If we're to include modern properties I'd love to see Tim Seeley's Revival turned into a live action series. Like a combination of The Walking Dead and The Leftovers TV series.

I don't really care for Live Action adaptations, always thought Animated was the best way to adapt comics.

On the Big Screen - I don't mind seeing an Aquaman Movie at all ... I would take it back to the 60's and make it somewhat nostalgic in style with people wearing period clothing of that era.

Didn't Ennis direct a short film himself, a war story of some kind? I seem to remember reading about that a year or two ago.
A lot of my favourite comics are of the sort I fear probably wouldn't translate well into any other medium - the New Gods, the Eternals, all the superhero stuff, ...
I could see something like Master of Kung Fu being turned into a first rate movie series, though. Maybe Steranko's Nick Fury, Agent of SHIELD.
But that brings up another issue: one thing I don't believe we've seen yet is a genuine effort to translate something of the visual effect of a really exceptionally well-drawn comic to the screen. Maybe the differences between the two media are too great for this to be feasible, but I'd like to see someone try, whether that means full animation or some other combination of techniques.
Would it be possible to make a Nick Fury film that captures the style of or at least is recognisably influenced by Steranko's artwork? The Thor movies certainly have not delivered this in regards to Kirby's artwork, though I know some fans disagree. I don't think any superhero film has even seriously tried anything like this yet. Spielberg's Tintin, which I haven't seen, might be the only attempt I've heard of, so far.
