Yes!!! Prophet counts because he appeared first in Youngblood #2.

Alas, Gwen Stacy didn't get her solo superhero series until 2015...

Nice choice as usual, Cei-U! . Already halfway there, I think. Do characters introduced in titles like Showcase, Marvel Premiere, Brave and the Bold and other "trial run" series counts as sources? Ooh, my favorite series is a spin-off of Amazing Fantasy by this logic.

Do characters introduced in titles like Showcase, Marvel Premiere, Brave and the Bold and other "trial run" series counts as sources? No, not if they were the feature character in their first appearance. For example, Lois Lane's solo series, which began in Showcase #9, would be eligible but Metamorpho's debut in B&B #57 would not. Clear enough?
Cei-U! I summon the difference!
Gotcha!

I guess this disqualifies Jonah Hex, who was the feature character in ASW #10

Can I get a ruling on Popeye? I’m assuming he qualifies but he spun out of a comic strip.

I’m reading this as comic strips being unwelcome this year. Yes, the strips are sitting this one out.
Cei-U! Sorry, strip aficianados!
Shucks. But good to know, thanks.

Do alternate timelines count? Its hard not to say Spider-Man 2099 is not a Spinoff, but techincally it would not meet the criteria you list, so I can definitely see it being excluded (same would apply to Ultimate line, perhaps). Hmmm, good question. I'm going to throw this open for group discussion. Should alternate versions of characters be counted as spin-offs? I'll make the call tomorrow, after you've all had a chance to have your say.
Cei-U! I summon the healthy debate!
Personally, I’d say any character who is an alternate version, with their own unique backstory, wouldn’t be a spin-off. I don’t know if many people back in the day used “spin-off” to describe Spider-Man 2099.

What about team books where the characters had originally appeared in their own titles?

I haven't started thinking about it seriously yet but I'm pretty sure I can come up with enough titles to fill the 12 days this year, so I hope to participate after sitting out last year.

I have a tentative twelve (the bad Italian version of The Dirty Dozen). I’m sure it will change numerous times in the next month.
