I may not be able explain this clearly. I'm wondering what successful comic book characters began as spin offs from another title?
Wolverine was my first thought. He began as a villain in the Hulk and somehow became a hero and then got his own book.
Of course, going further back, Jimmy Olsen and Lois Lane were characters in Superman who go their own long running titles.
What other supporting characters were popular enough to get their own titles?

but the whole Superman family shared a spin off title (Batman Family too).
Secret Society of SuperVillains too.
and there are multiple spin offs from LSH -- including L.E.G.I.O.N., Legionaries, and the one featuring the heroes of Llalor. . what was that one called again. . was that "the Wanderers" ??

Deadpool. Started out as a villain mercenary in the dying days of the New Mutants title, then became an anti-hero over time, to carrying multiple solo titles and starring in his own movie.
The Punisher, obviously, from his debut in ASM 129 to carrying three solo titles at one point and being the subject of two solo films.

Off the top of my head: Superboy, Supergirl. Jughead. Betty and Veronica. Uncle Scrooge. Tubby. Captain Marvel, Jr. Mary Marvel. The Black Panther. The Inhumans. The Silver Surfer. Silver Age Ka-Zar. Moon Knight (originally a Werewolf By Night villain). Ms. Marvel. She-Hulk. The Cave Kids.

Technically, wouldn't all of the Legion of Super-heroes spinoffs be spinoffs of a spinoffs?

I think Legion is probably the longest running... if you call that a spinoff (not sure I would.. it's more just a feature that started in another book)... maybe Uncle Scrooge (depends how you count, probably)
No one mentioned Robin yet.. he's gotta be up there with Punisher and Wolverine I think.
Then there's the X-books (X-Factor, New Mutants, X-Force, Gen X, etc).. the Avengers books (West Coast, New, etc).... Justice League spinoffs (Europe, Task Force, Extreme). I'd call Thunderbolts a spin off as well.
If you're talking solos only. Deadpool is probably the most popular atm.

More like the stereotypical Marvel "misunderstanding."

Moon Knight began his career as a villain in Werewolf by Night, but was retconned as a hero when it became obvious fans liked him.
