I've got a tentative 12. This assignment is well outside my wheelhouse, so I keep wondering if I'm missing some obvious, better choices, but this just might do.
Tentatively, here are some stats:
5 DC 2 Marvel 5 other
3 couples primarily selected for their relationship in the 1970s 5 couples primarily selected for their relationship in the 1980s 2 couples primarily selected for their relationship in the 1990s 1 couple primarily selected for their relationship in the 2000s 1 couple that I couldn't assign to a specific decade
7 couples where the male is the primary character 4 couples where the male and female are equal in status 1 male-male couple, both equal in status 0 couples where the female is the primary character 0 female-female couples (though two were close runner-ups).

Of my tentative 12, only one of them is outside DC and Marvel.
I have read other publishers, but not on a consistent enough basis to warrant having a view on a couple’s romance.
But I am glad that, at least as of writing, one of them is outside the Big Two.

Or Krazy Kat and Ignatz.
That's one that occurred to me almost immediately, and it is particularly interesting given Krazy's rather ambiguous gender identity. However, the romance - such as it is - is based on a pretty abusive relationship. And anyway, I always thought of the core relationship in that comic as a love triangle, with Offisa Pupp as the third member.

I think I could do 12 outside Marvel and DC just by doing comic strip couples and never touch comic book couples at all. But mine will be a mix of comic strip and comic book couples I'd imagine. My brainstorm list had a comic book couple as the first I jotted down (non-Marvel/DC) followed by about 10-12 comic strip couples before I even started thinking about super-hero couples.
I have no idea how I will pare it down or order it yet, but I've got two weeks before I have to decide what my first entry will be. I have a feeling it might come down to which couple do I want to feature today as I do my entries.

I have a tentative list but it’s so DC heavy is ludicrous.
A guy could almost do a list just from Terry & the Pirates.

I thought quite a bit about newspaper strip couples but since I read those sporadically several came to mind. Problem was they just didn't STAND OUT in my thoughts so they are part of the 40+ list and not really near the top as FAVORITES for me.
Could easily go 12 Marvel or 12 Legion of Super Heroes but wanted to really look at memorable and significant couples I really enjoyed reading and were more thought out than thrown together. No Doctor Octopus and Aunt May stuff.

Yeah, mine was pretty DC heavy myself. Though my "also rans" are more diverse, there are so many pairings from excellent DC stories that I just couldn't ignore them.

I have a bunch I would have included, but they are too recent to use for the list. The cut off takes a number of diverse couples I really enjoy reading about off of my list as they just haven't been around long enough for consideration. I might through some of them out there in my near miss posts.

I think the biggest struggle I'm having with this list is those relationships that seemed ideal to me when I first read them, but which (upon further consideration) are really one-sided, with one partner doing all the giving/understanding/caring, and the other simply doing the taking. Comics certainly idealize those relationships a whole lot. I left three one-sided relationships on my list, but, in each case, these are broken relationships, and that was sort of the point. We can still be invested in relationships that we know aren't working. It's the ones that masquerade as healthy and ideal when they aren't that bug me.

You hit it on the head for what my personal choices are leaning into. I know there is likely to be some obvious choices turn up so I was racking my head to focus upon those pairings which were NOT so one sided or unhealthy. Mileage may vary for every reader. Hoping that a few of my choices may delight...

This is definitely not something I've ever thought about... romance/relationships in literature and comics are not my thing... I generally stay away from them. I can't even think of one from a comic strip other than a couple golden age-y ones... I mostly read gag strips.. I meanm, I guess some of them have couples in them, but... not sure.
I got a few though, for sure. #1 was easy. Got at least 2 I suspect will be unique. still need a couple more though.

I've got my list more or less finalized, and I'm pretty happy with it. TOUGH assignment, but extremely rewarding. One more issue I've had to contend with in working this out is that most of the relationships I've selected really didn't have the steam to "go the distance" long term. Maybe four of these could have resulted in happy/healthy marriages, but it's very possible to love short and intense romances that have no future, admiring them for what they are while also recognizing what they are not. Anyway, my top two were absolutely totally healthy and admirable long-term relationships (because that's what I admire most when discussing love), but I have an extremely soft spot for the rest too.
