Original to comic books, so not Casper or Donald Duck (animation) nor Sad Sack or Little Lulu (magazines/newspapers) for example, but not super-heroes. Who would be in the hall of fame based on longevity or appearing in other media after starting in comics?
I'd like to nominate:
Sheena, Queen Of The Jungle. She had a '50s tv series, an '80s movie, and ran in comics solidly from the late 1930s to middle 1950s, plus some less successful revivals, mostly reprints. A lot of people still remember the name and basic character.

Men in Black... started as a comic and spawned the hit movies. the Crypt Keeper, EC's horror host live on in movies, TV and animation long after EC ceased publication and was front and center in several comic revivals over the years as well. Tintin....animation, movies, and books that have millions of copies in print worldwide... -M

I'd say the Bone crew too simply because of the ubiquitousness of the Bone books in Scholastic programs there are 2-3 generations of kids who will always know these characters and the movie in the works could cement those characters place in pop culture permanently. -M

Uncle Scrooge-created in the comics, brought a world of fun and adventure to generations, with new ones discovering it, via the revived Duck Tales.
Maggie and Hopey-Love & Rockets best-known characters; real women with real feelings, in engaging stories.
Richie Rich-the Poor Little Rich Boy went from back-up in Little Dot, to carrier of the company in short order. It presented a world of imagination, fun, and good moral lesson, in a package that delighted generations, leading to cartoons, film and the recent tv series.

I think Archie might be the number one for longevity and success in other media (I had/have all the Archies LPs except the first one, just a CD for that). I actually didn't realize Men In Black started in comics! Uncle Scrooge is a very important original too! I'm ashamed I didn't think of him as I did mention Donald. I liked Scamp comics but I believe he actually briefly appeared at the very end of Lady & the Tramp, so... Already quite a few well known and international (Tintin) characters to have started in comic books. How about Pogo? He started in Dell's Animal Comics if I'm remembering things straight. I had an Eclipse book collecting a lot of the early appearances, some with a kid named Bumbazine, and maybe Albert Alligator was there too.

I used a Pogo panel for my avatar here for quite some time (and still do at another comic site) However, I don't think Pogo is as well known today as he has been in the past, so the staying power of the property might be in question. -M

Hmm, not a big fan myself, but Rick Grimes and the characters from the Walking Dead might be the current most well known non-super-hero characters originating in comics in mass pop culture.

Pogo might have fallen lower down on the fame list I guess along with Sheena. I wonder if Sgt. Rock is well known either anymore?

He may not meet the criteria of "longevity in other media" but has a good combination of "longevity in comic books, and known worldwide today":
